In Situ Monitoring of CHO Cell Culture Medium Using Near-Infrared Spectroscopy

Abstract

This study was performed to demonstrate the application of near-infrared spectroscopic analysis to CHO cell culture medium. A bioreactor was analyzed in situ during a CHO cell culture in order to effectively monitor nutrient levels and ammonia production. Spectra were collected over the 800 nm to 2100 nm wavelength range using fiber optic cables and a transmission probe with a 4 mm gap/pathlength. The glucose, ammonia, titer, methionine, lactate, glutamate and glutamine levels were monitored spectrally and regressed against lab data. Prediction models were developed that predicted trend plots of the culture media constituents with time. The trend plots show smooth trends that pass through the reference data, indicating good prediction model performance.
