AstraZeneca’s Imfinzi-Lynparza Combination Misses Overall Survival Goal in Ovarian Cancer Trial

AstraZeneca reported disappointing results from its Phase 3 DUO-O trial, revealing that the combination of Imfinzi (durvalumab) and Lynparza (olaparib) failed to meet overall survival (OS) goals in patients with newly diagnosed advanced ovarian cancer. The study, presented at the ESMO 2025 Congress, built on earlier findings showing a 38% improvement in progression-free survival (PFS), but those gains did not translate into a statistically significant survival advantage.

The triple regimen of Imfinzi, Lynparza, and Avastin (bevacizumab) alongside standard chemotherapy showed encouraging tumor control benefits but fell short on key secondary endpoints. The trial involved more than 1,200 participants without BRCA mutations, a group representing a major unmet need in ovarian cancer therapy.

AstraZeneca stated that the data will inform ongoing discussions with regulators and future studies. Dr. Susan Galbraith, Executive Vice President of Oncology R&D, emphasized that despite the OS shortfall, the combination’s PFS benefit reaffirms the potential of pairing immunotherapies with PARP inhibition for select subgroups.
