Addex Therapeutics has regained full global rights to GABAB positive allosteric modulator assets discovered through its research collaboration with Indivior UK, including a development candidate for substance use disorders.
The return follows the termination of the collaboration and Indivior’s decision to reorganize its research and development activities. Addex also cited the planned merger of Indivior’s parent company with Supernus Pharmaceuticals.
Addex now owns the returned candidate and can pursue indications beyond those covered by its former agreement with Indivior. The substance-use-disorder candidate has completed studies needed before an investigational new drug application, Addex said.
The returned assets will join Addex’s wholly owned GABAB modulator program for chronic cough. That candidate is ready to begin studies needed to support an investigational new drug application. Addex said it may seek partners for the programs or develop them further before pursuing a partnership.
GABAB receptors help regulate nerve-cell activity. Positive allosteric modulators are designed to enhance the receptors’ response to GABA, a neurotransmitter, by binding at a site distinct from GABA’s usual binding site. The approach differs from baclofen, an approved drug that directly activates the receptor and is used to treat spasticity.
Addex identified substance use disorders and chronic cough as the focus of its current GABAB candidates. It also cited pain, overactive bladder and neurodevelopmental disorders as areas in which the receptor pathway may be studied. The release did not report clinical results for the returned candidate.
The Geneva-based company develops small-molecule drugs for neurological conditions. Its separate lead candidate, dipraglurant, is ready for Phase 2 development and is under evaluation for recovery after stroke or traumatic brain injury.
