Endosafe® Trillium™ recombinant cascade reagent (rCR) cartridges are designed for bacterial endotoxin testing (BET) without using animal-derived materials. The product provides rapid, quantitative endotoxin detection that supports sustainability goals and regulatory compliance in pharmaceutical and medical device manufacturing.

Features and Benefits

Quality control microbiology labs face two main challenges. First, traditional gel-clot or plate-based endotoxin tests are slow, manual, and often produce inconsistent results. Second, labs are under pressure to adopt more sustainable practices—such as replacing animal-based methods—with options that align with the 3Rs: Replacement, Reduction, and Refinement.

The Trillium rCR formulation within the cartridge technology detects and quantifies natural environmental endotoxins by simulating the LAL enzymatic cascade through an optimized formulation and composition of three recombinant proteins (recombinant Factor C, recombinant Factor B, and recombinant proclotting enzyme) and a specific concentration of key components. Cartridges come pre-loaded with all necessary reagents, reducing hands-on steps and minimizing variability.

It’s fully compatible with Charles River’s existing cartridge instruments and software, allowing labs to transition to recombinant reagents without new equipment or retraining. The assay delivers results across a quantitative range of 1 to 0.01 EU/mL, and 5 to 0.05 EU/mL to support batch testing and routine workflows. The formulation removes the risk of 1,3-β-D-glucan interference, helping reduce false positives.

For QC teams aiming to modernize their workflows with an animal-free method, Trillium improves reliability, reduces waste, and integrates into existing operations without added friction.

Applications

Trillium cartridges are used in raw material, in-process, and final-release testing across thousands of products. They support companies working to increase efficiency and accelerate timelines while moving toward more sustainable testing practices. Approved product applications include pharmaceutical products, biologicals, nuclear medicine samples, dialysis samples, pharmaceutical water systems, stem cell materials, cleaning validations, medical devices, and compounded products.
