Merck’s oral cancer drug Welireg (belzutifan) has posted highly positive results in two pivotal Phase 3 clinical trials targeting advanced and early-stage kidney cancer, marking a milestone in renal cell carcinoma therapy advancement. The first trial evaluated Welireg in combination with Lenvima (lenvatinib) for patients with advanced renal cell carcinoma (RCC) who had failed previous immunotherapy. This combination achieved statistically significant improvement in progression-free survival, delaying disease progression compared to current standard treatments.
In a separate study, Welireg was paired with Merck’s blockbuster immunotherapy Keytruda (pembrolizumab) in patients with clear cell RCC who had undergone nephrectomy, the surgical removal of the kidney. Here, the combo demonstrated a clinically meaningful boost in disease-free survival, outperforming Keytruda alone, the longstanding standard for adjuvant therapy in post-surgical kidney cancer patients.
Welireg, a first-in-class HIF-2α inhibitor, is now positioned as a key asset in Merck’s kidney cancer franchise as patent expiration for Keytruda nears. If these trial results support expanded approvals, analysts estimate the market for Welireg in new indications could rise to $6 billion annually, tripling its current revenue projections.
